Kinase Target-Based Whole Cell Precision Screening Library.

This cell-based assay library is built from mouse Ba/F3 cells that are natively dependent on IL-3 for growth. After transformation through retroviral infection to express constitutive human recombinant kinase proteins, the cytokine dependence is switched to the activity of the transgenic recombinant kinases for cell survival and proliferation.

With these modified Ba/F3 cells, specific kinase inhibitors will inhibit the activity of the introduced recombinant kinases, which in turn subject these cells to programmed cell death. Since the inhibition of kinase activity leads to directly proportional decrease in cell viability, this library proves to be an indispensable tool for studying the potency and specificity of kinase inhibitors.
